Advanced Analytical Chemistry: Lecture 1

We’re going to begin a new series today. This covers some analytical chemistry, which deals with instrumentation. Our first lecture costs 29 minutes. The audience appears to be graduate students.

Two immiscible liquids. Perhaps hexane and water. Put in some compounds we’d like to seperate. Put them in at low concentration for a linear chromotography. Benzene would stay with the hexane.

A seperatory funnel can only seperate two phases. Can’t get 100% purity. To do a perfect amount of seperation, you need infinity entropy.

Resonance or entropy is usually the answer whenever something really bothers you.

We think of the number of plates in a chromatography column. It’s a continious process that we think of discontiniously.

Partition factor = K =
K = products/reactants

Moles = concentration x volume

Phase ratio = Xa organic/ Xa polar

Retention factor = Xa/X total

RF tells you where something is because it shows the fraction in either phase.

The solute in the stationary phase is a reactant, in the mobile phase is a reactant.

The stronger the interactions between the polar analyte and the polar stationary phase (relative to the mobile phase) the longer the elution time.
